Cell reprogramming can be used to short-circuit the cancer-immunity cycle Fábio F. Rosa Cancer remains one of the greatest challenges to modern medicine, affecting one in three people worldwide. In the past few decades, immunotherapies such as immune checkpoint blockers (ICB) and chimeric antigen receptor (CAR) T cell therapies have transformed cancer treatment by delivering durable responses and unprecedented long-term survival. However, these therapies benefit only a minority of patients and often lose efficacy as tumors progress, become heterogeneous and immunosuppressed, and evade immune detection. Addressing these challenges requires new strategies capable of restoring the immunogenicity of established tumors and eliciting durable, tumor-agnostic immune responses to bring the benefits of cancer immunotherapy to every patient. …
