Richardson Long interspersed nuclear element–1 (LINE-1 or L1) is a type of mobile DNA element that can insert itself into genes and disrupt their func- tion. The human genome contains ~500,000 L1 copies, most of which are inactive (1). Although ongoing L1 retrotransposition in humans was recognized nearly four decades ago (2), the extent to which this activ- ity contributes to human disease is unclear.
