本文讨论了周等人开发的一种名为CRYSTAL的纳米颗粒,该颗粒将STING激动剂递送至肿瘤,在动物模型中引发强效抗肿瘤免疫反应且毒性有限。
原文开头
STING agonist nanoparticles trigger potent antitumor immunity in mice and rabbits Rick Liao and Darrell J. Irvine The stimulator of interferon genes (STING) protein is a component of the innate immune system. The STING pathway detects bacterial cyclic dinucleotides (CDNs) and double-stranded DNA in the cytosol of diverse cell types, which drives a robust immune response. This has raised the possibility of activating STING therapeutically to stimulate antitumor immunity. However, it has been challenging to develop STING activators that can target metastatic tumors, which are widespread across the body, without causing intolerable body-wide inflammation.
