mammalian vitreous body collagen fibrils In this work, we present the high- resolution structure of native col- lagen fibrils isolated from porcine vitreous humor. Each 67- nm peri- odic unit contains triple helices of collagens II, V/XI, and IX, associated with the noncollagenous glycoprotein opticin. We observed that ex- tensive O- linked glycosylation, consisting of glucosylgalactosyl modi- fications on Hyl residues in the - Gly- X- Hyl- sequence pattern, stabilizes interhelical packing in the fibril. Our results uncover the molecular architecture and assembly principles of native vitreous collagen fibrils, provide insights into the pathogenesis of collagen- related diseases, and establish a structural framework for designing and engineering collagen- based biomaterials. …
