INTRODUCTION: The human X chromosome has two unusual properties. First, in XX individuals, X-chromosome inactivation (XCI) silences one X chromosome, ensuring that X-linked gene dosage is equivalent to that of XY individuals. Second, ∼30% of the X chromosome is composed of long interspersed element-1 (LINE-1 or L1) retrotransposons, a type of mobile DNA, and this percentage is double the autosome average. In 1998, Mary Lyon connected these features and proposed that L1 sequences on the X chromosome may facilitate XCI and accumulate due to positive selection. …
