structure determination of small proteins by cryo- EM We have developed a laser phase plate (LPP) and demonstrated that it has superior stability while also showing negligible signal attenu- ation (14–17) and decoherence from thermal effects (18). Instead of placing a material phase shifter into the beam path, it induces the phase shift by stimulated Compton scattering (19) with a continuous light intensity of ~400 GW/cm2, the highest ever demonstrated. A 12- W, 1064- nm laser beam is built up to 80 kW circulating power in a Fabry- Pérot cavity, where it is focused to a waist of 6.7 μm (1/e2 intensity radius) (16). …
