Silverman, Louis- Charles Campeau, Ralph Pantophlet, Robert Britton* INTRODUCTION: Nucleosides, composed of a carbohydrate core linked to a heterocyclic nucleobase, are the fundamental building blocks of DNA and RNA and play essential roles in the metabolic and reproductive processes of all living organisms and viruses. Nucleoside analogs (NAs) constitute a major drug class with a critical role in the treatment of cancer and viral infec- tions. Despite the existence of >30 NA drugs approved for clinical use, their chemical synthesis remains challenging, constraining structural diversity and limiting the discovery of new therapeutics. …
