Yet glycan structural biology remains severely hampered by chemical complexity, conformational flexibility, and nontemplated synthesis. The CryoSeek strategy, which represents a structure- first paradigm for discovery, has yielded dozens of high- order structures of glycans in the form of glycofibrils. However, the ~3.0- Å resolution of these glycans precludes reliable sugar assignment. …
