Sophie A. Hanina et al. INTRODUCTION: Chimeric antigen receptor (CAR) T cells are living drugs that are programmed to eliminate cancer cells. These cell therapies are effective in hematological malignancies but currently fail in solid tumors. One challenging obstacle to the success of CAR therapy owes to the inconsistent expression of target antigen across the population of tumor cells, which results in incomplete tumor elimination and overall disease progression. RATIONALE: The properties of an ideal target include (i) expression on all tumor cells, (ii) restricted expression on dispensable normal cells, and (iii) common to many tumor types. CD70 is potentially one such target, fulfilling criteria (ii) and (iii), but its expression pattern (i) is typically heterogeneous in solid tumors. …
